摘要
为研究血啉甲醚(HMME)与小鼠血浆的蛋白结合率及HMME与血浆蛋白的结合参数,用平衡透析法测定蛋白结合率,HMME浓度用标准曲线法求出。HMME血浆浓度在194μg/ml~2976μg/ml时血浆蛋白结合率为935%~763%,血浆蛋白结合参数:最大结合力(β)为87×10-6mol/g;解离常数(Kdp)为424×10-5mol/L;结合常数(K)为284×104L/mol;结合部位数(N)为1492。表明HMME与小鼠血浆蛋白具有单一类型的结合部位,药物血浆浓度升高结合率下降。
o elucidate the binding of hematoporphyrin monomethyl ether (HMME) to plasma protein of mice and evaluate the binding parameters, the binding rate of HMME to plasma protein was determined by equilibrium dialysis method. Concentration of HMME was assayed by standard curve method. The binding rate of HMME to plasma protein was found to be 93.5%-76.3% varying with the concentration of HMME in plasma from 19.4 μg/ml-297.6 μg/ml. The binding parameters were 8.7×10^-6mol/g for β; 4.24×10^-5mol/L for Kdp; 2.84×10~4 L/mol for K and 1.492 for N. There were singletyped binding sites for HMME to plasma protein of mice. Moreover plasma concentration of HMME was inversely proportional to the binding rate.
