摘要
以桃品种曙光、甜桃王和沙红的幼胚胚轴为试材,对影响桃胚轴产生愈伤及再生不定芽的培养基、切口部位和继代次数等因子进行研究,结果表明:(1)诱导桃胚轴产生愈伤和再生不定芽的培养基相同,曙光、甜桃王为LP+TDZ3.0mg·L-1+BA1.0mg·L-1+NAA1.0mg·L-1,其愈伤诱导率达100%,不定芽再生率分别达24.44%、55.56%;沙红为MS+TDZ4.0mg·L-1+BA1.0mg·L-1+NAA0.1mg·L-1,愈伤诱导率达100%,不定芽再生率达46.67%。(2)近子叶端切口产生的不定芽数显著高于远子叶端,曙光、甜桃王和沙红的平均不定芽数分别达6.33,9.67和8.00个。(3)随着继代次数的增加,愈伤的形态变化差异较大,且再生不定芽的能力也存在明显差异。
Experiment was conducted with Shuguang,Tiantaowang and Shahong(Amygdalus persical L.) for studying the effects of different elements in medium,cut place and subculture time on inducing callus and regeneration of adventitious shoots of embryonic axis.Results showed:(1)The best medium for both callus inducing and adventitious shoot regeneration from emhryonic axis of shucuang,Tiantaowang and Shahong was same;For Shuguang and Tiantaowang on medium of LP+ TDZ 3.0 mg·L^-1+BA 1.0 mg·L^-1+NAA 1.0 mg·L^-1,the ratio of their callus was 100%,the ratio of adventitious shoots was 24.44% and 55.56% respectively;for Shahong on medium of MS+TDZ 4.0 mg·L^-1+BA 1.0 mg·L^-1+NAA 0.1 mg·L^-1,the ratio of callus reached 100%,while the ratio of adventitious shoots was 46.67%.(2) The adventitious shoots regenerated from near cotyledon end were significantly more than that from far end of these three cultivars,their average adventitious buds regenerated were 6.33,9.67 and 8.00.(3) Along with the subculture times increasing,there appeared great difference on the morphology of callus,and the ability of regeneration of adventitious shoots was obviously different as well.
