刺五加不同部位的红外光谱分析与鉴定 被引量:3

Analysis and identification different parts from Acanthopanax senticosus by Fourier Transform Infrared Spectroscopy

摘要 目的对刺五加根、茎、叶原药材及总苷提取物的红外光谱图进行整体分析。方法采用红外光谱三级宏观指纹鉴定的方法。结果原药材谱图显示,根和茎谱图较为相似,主要体现在草酸钙和淀粉特征峰,而叶的草酸钙特征峰几乎消失,淀粉特征峰的峰形也变得不明显。总苷提取物的一维谱图显示根、茎、叶在1602、1514、1452 (苯环骨架振动)、1271 cm^(-1)(=C—0)附近均有特征吸收,因此,推测3者共有的成分为酚苷类化合物。二阶导数谱图给出叶在1656 cm^(-1)(黄酮的C=0)附近特征峰明显强于根和茎,说明叶中黄酮类成分高于根和茎。二维谱图在1350~1700 cm^(-1)波段根和茎均体现苯环骨架振动的5个自动峰1636、1600、1521、1462、1453 cm^(-1),叶除了苯环骨架振动峰外还出现1656 cm^(-1)黄酮类成分的C=O振动,进一步证实叶中黄酮类成分高于根和茎。结论初步建立了刺五加的宏观质量控制标准,提供了宏观质量控制的重要参考数据和评价依据。 Objective To analyse the multi-steps IR macro-fingerprint ot raw radix, stem, leaf, anti total glycoside extract of Acanthopanax senticosus. Methods By Multi-steps IR macro-fingerprint Method. Results The spectra of raw medicinal materials show that the spectra of radix and stem are similar and mainly show characteristic peaks of calcium oxalate and starch, whereas the characteristic peaks of calcium oxalate of leaf almost disappears and its shape of characteristic peak of starch also gets unconspicuous. FT-IR spectra of total glycoside extract of radix, stem, and leaf present characteristic peaks at 1 602, 1 514, 1 452 (vibration of phenyl framework)and 1 271 cm^-1(=C--O),therefore, their mutual component is the compound of phenolic glycoside. Through observing second derivative IR spectra of total glycoside extract of different parts of Acanthopanax senticosus Harms, the characteristic peak of leaf is stronger than that of radix and stem at 1 656 cm^-1 (flavone C=O), it proves the quantity of component of flavone in the leaf is higher than that in the radix and stem. In two-dimensional correlation spectra, the radix and stem both have five automatic peaks (vibration of phenyl framework) at 1 350-- 1 700 cm^-1, whereas the leaf still shows another automatic peak at 1 656 cm^-1(flavone C=O), it further proves the quantity of component of flavone in the leaf is higher than that in the radix and stem. Thus it can be seen IR macro-fingerprint technique can provide plentiful holistic information and hold general quality of Acanthopanax senticosus Harms. Conclusion The multi-steps IR macro-fingerprint method was demonstrated to be a rapid, simple, and reliable method on the quality control of Traditional Chinese Medicine.
