摘要
目的研究微波辐照对小鼠睾丸的损伤效应。方法采用平均功率密度为65mW/cm2和90mW/cm2微波辐照小鼠15min,采用HE染色和透射电镜方法观察小鼠睾丸组织的改变,用免疫组化方法观察计数睾丸内Caspase-3阳性细胞的表达情况。结果微波急性辐照后12h,小鼠睾丸组织在光镜和电镜水平均出现损伤性改变,但65mW/cm2及90mW/cm2两个剂量组之间在形态学改变方面差异无统计学意义;辐照后0.5h Caspase-3蛋白阳性细胞数开始出现升高,24h达到高峰,其中90mW/cm2组升高幅度高于65mW/cm2组。结论微波对小鼠睾丸组织存在明显损伤效应,并且能诱导凋亡相关因子Caspase-3表达增强,诱导效应存在着时效和量效依赖关系。
Objective To investigate the damaging effect of mice testis exposed to microwave irradiation. Methods The mice were irradiated by microwave(mean power 90mW/cm^2 or 65mW/cm^2) for 15min respectively. The histological changes of mice testis was determined by HE stain and transmission electron microscope, and the expression of Caspase-3 protein was determined by immunohistochemistry. Results The damaging effect of testis exposed to microwave via transmission electron microscope was observed,and these damages reached peak at 12h,but there was not significant difference between 90mW/cm^2 group and 65mW/cm^2 group;it reached peak at 12h. The expression of Caspase-3 protein increased after 0.5h and reached peak at 24h,and the increasing in microwave of 65mW/cm^2 group,it reached peak at 4h and recovered no difference with control group at 12h. The increasing extent of 90roW/cm^2 group was higher than that of 65mW/cm^2 group. Conclusion Microwave could damage mice testis with up-regulated significantly the Caspase-3 protein and the up-regulating may be dependent on the time and dose of irradiation.
