摘要
目的:探讨去甲斑蝥素(NCTD)对人乳腺癌细胞血管内皮生长因子(VEGF)表达的影响及作用机制。方法:应用四甲基偶氮唑盐(MTT)法检测去甲斑蝥素对人乳腺癌细胞系MDA-MB-231和MCF-7细胞增殖的影响;应用酶联免疫吸附试验(ELISA)检测VGEF蛋白的分泌水平;采用半定量逆转录-聚合酶链反应(RT-PCR)技术检测VEGF mRNA表达水平。结果:去甲斑蝥素对DA-MB-231和MCF-7细胞均有明显增殖抑制效应。去甲斑蝥素降低了VEGF分泌水平、VEGF mRNA的表达水平。并呈剂量-时间效应关系。结论:去甲斑蝥素能抑制人类乳腺癌细胞增殖和细胞VEGF mRNA的表达。
Objective: To assess the effects of norcantharidin on VEGF mRNA level in human breast cancer cell lines. Methods: MDA-MB-231 cells and MCF -7 cells were used as the study objects. MTr assay was used to detech the effect of NCTD on the cells viability. ELISA was used to determine the protein level of VEGF. Breast cancer cells were treated with NCTD for 24 -~ 72h, RT - PCR was used to detect the level of VEGF mRNA expression. Results : NCTD had visible inhibitory effect on cell viability. NCTD reduced the secretion of VEGF and inhibited the level of VEGF mRNA expression. Conclusion: NCTD can inhibit cell viability and the level of VEGF mRNA expression in breast cancer ceU.
