摘要
目的:通过检测血管内皮生长因子(VEGF)和间质金属蛋白-酶2(MMP-2)及金属蛋白酶组织抑制剂-2(TIMP-2)在肾细胞癌(RCC)中的表达情况以探讨它们在肾癌的发生、侵袭和转移过程中的作用。方法:用免疫组织化学方法检测40例RCC石蜡标本中MMP-2、TIMP-2和VEGF的蛋白表达情况,并进行统计学分析。结果:MMP-2和VEGF在RCC中表达率明显升高,但不同病理分级、临床分期间差异无显著性(P>0.05);淋巴结转移阳性组MMP-2和VEGF的表达率均明显高于淋巴结转移阴性组,两者之间存在显著性差异(P<0.05);TIMP-2与MMP-2表达存在负相关性,TIMP-2对MMP-2有一定拮抗作用。结论:MMP-2与TIMP-2平衡状态的改变,很可能与RCC的侵袭和转移密切相关。VEGF可作为判断RCC淋巴结转移和临床预后的生物学指标之一。
Objective: To investigate the expression of vascular endothelial growth factor (VEGF), matrix metalloproteinases-2(MMP-2)and its inhibitor tissue inhibitor of metalloproteinase-2(TIMP- 2) in renal cell carcinoma(RCC). Methods: Protein expression of MMP-2 ,TIMP-2 and VEGF in 40 RCC tissues was examined by immunohistochemical assay. Results: MMP-2 and VEGF showed an overexpression rate in RCC, but there were no significant differences among different pathological grades and clinical stages. The expression of MMP-2 and VEGF was significantly different between the metastasis group and the non-metastasis group (P〈0. 05). A negative correlation was detected between MMP-2 and TIMP-2. TIMP-2 has a inhibitory action on MMP-2. Conclusion: Overexpression of MMP-2 appears to have prognostic significance with respect to the invasion and metastasis of RCC. The effect of MMP-2 can be restrained selectively by TIMP-2. The balance between MMP-2 and TIMP-2 shows an important role in the development and invasion of RCC. VEGF may also serve as a predictor of clinical prognosis and lymph node metastasis.
