摘要
目的:通过观察电针对PD大鼠的行为学指标的影响及其酪氨酸羟化酶mRNA表达,探讨电针对帕金森病大鼠的治疗效应和电针治疗帕金森病的可能机理。方法:通过多位点纹状体内注入6-OHDA制作PD大鼠模型。分组进行电针治疗,于治疗前、后,分别检测各组实验大鼠的旋转行为。在此基础上,采用逆转录聚合酶链反应检测各组实验大鼠中脑黑质中TH mRNA的蛋白表达,进而初步探讨电针对帕金森病治疗作用的部分机理。结果:对各组实验大鼠行为学检测显示,与模型对照组比较,电针可以有效改善帕金森病大鼠的旋转行为。通过检测各组实验大鼠中脑黑质中TH mRNA的蛋白表达情况,证实电针头穴能使帕金森病大鼠中脑酪氨酸羟化酶基因表达增加。结论:电针对帕金森病模型大鼠的旋转行为具有良好的改善效果,其机制可能是通过增加中脑TH mRNA的蛋白表达,以控制帕金森病进一步的发展。
Objective: Observing the effect of the electroacupuncture on the rotational behavior, and to discuss the partial possible mechanism by observing the expression of THmRNA in PD rats. Methods:The rat model of PD was made by injecting the 6 - hydroxy dopamine into the striatum from four points. Before and after treatment, experimental rats were measured the rotational behavior. To observe therapeutical effect of electro - acupuncture on rat model of PD according to the rotational behavior. In addtion, the expression of THmRNA was detected by RT - PCR. Results : Contrasted with the control group, expression of THmRNA was detected, which confirmed that EA could increase expression of THmRNA and improve the rotational behavior. Conclusion:EA could improve the rotational behavior of rat model of PD, and control the development of PD by increasing expression of THmRNA.
