微透析法研究磷酸川芎嗪体外血浆蛋白结合率 被引量:5

Determination of Plasma Protein Binding Rate of Tetramethylpyrazine Phosphate in Vitro by Using Microdialysis Method

摘要 目的测定磷酸川芎嗪的血浆蛋白结合率。方法采用微透析法测定磷酸川芎嗪的血浆蛋白结合率,用反透析法进行探针回收率校正。血浆蛋白结合率用f=(ρ0-ρf)/ρ0计算。结果磷酸川芎嗪与牛血清白蛋白、人血清白蛋白和人血浆、大鼠血浆的蛋白结合率分别为(79.51±1.66)%、(68.34±1.43)%、(56.17±1.44)%和(70.97±2.19)%。结论磷酸川芎嗪与血浆蛋白具有中等强度的结合,采用微透析进行血浆蛋白结合率研究可行。 OBJECTIVE To determine the plasma protein binding rate of tetramethylpyrazine phosphate in vitro by microdialysis method. METHODS The binding rate of tetrarnethylpyrazine phosphate was determined by microdialysis probe as sampling tools and retrodialysis method as calibration. The plasma protein binding rate was calculated using the equation: f=(Po-Pf)/ P0. RESULTS The binding rate was kept relatively stable in the concentration range. The plasma protein binding rate of tetramethylpyrazine phosphate with BSA, HAS, human plasma and rat plasma were (79.51±1.66) %, (68.34±1.43) %, (56.17±1.44)% and (70.97±2.19) %, respectively. CONCLUSION It is feasible that the plasma protein binding rate can be determined by microdialysis. The strength of binding rate of tetramethylpyrazine phosphate with plasma protein is middle and the tetramethylpyrazine phosphate can be determined.
