摘要
前言乙肝病毒c基因的克隆和表达的研究,国内外近年来有多篇报道。近有学者提出乙肝核心抗原(HBcAg)的前c区(PreC)可能为HBcAg分泌所必须,它使后者嵌合在肝细胞膜上成为靶抗原,产生免疫反应而造成肝细胞的损伤。本文将为HBcAg编码的c基因分别和载体pUR222及pⅠN-ⅡC重组得到重组质粒pHW202和pDC022。
Using DNA recombinant technology, we constructed recombinant plasmids with DNA fragment carrying HBc gene and vectors: pUR222 and pINIIC. Recombinants was transformed to E. coli and HBc gene expressed at high level in the cytoplasmic and membrane fractions of E. coli respectively. The E. coli derived HBcAg have been used to detect anti-HBc in 265 sera samples. The results correlated very well with that obtained by using other ELISA or RIA kit currently used in China.
