摘要
目的观察雷公藤甲素对胶原诱导型关节炎(CIA)模型小鼠骨与关节破坏的影响。方法Ⅱ型胶原尾根皮下2次免疫诱导DBA/1 小鼠建立CIA模型,第1次免疫后14~42 d,各给药组分别灌服雷公藤甲素(8.18、 16.36、32.72 μg/kg)和甲氨蝶吟(0.1 mg/kg),定期观察临床症状和发病情况,42 d取材,观察组织病理学和放射学变化,并用抗酒石酸酸性磷酸酶(TRAP)对破骨细胞特异性染色后进行TRAP阳性细胞计数。结果雷公藤甲素能显著降低CIA小鼠的临床积分和发病率,减轻关节滑膜、软骨和骨的组织病理学变化及X光片的骨破坏程度,改善关节间隙,增加膝关节和腰椎骨密度,降低膝关节的骨髓腔和炎性肉芽组织中TRAP阳性细胞的数量。结论雷公藤甲素对CIA小鼠的关节炎症和骨破坏具有一定的抑制作用。
Objective To observe the effect of triptolide on destruction of bone and joint of collageninduced arthritis (CIA) mouse. Methods Female DBA/1 mice were double immunized at the base of the tail with bovine type Ⅱ collagen (CⅡ). CIA mice were randomly divided into model group, methotrexate-treated group and 8.18, 16.36, 32.72 μg/kg triptolide-treated groups. Performance of arthritis were observed regularly. Combined with radiological, histological methods were used, and the number of osteoclasts in bone was evaluated and analyzed with TRAP stainning. Results The clinical score and arthritis incidence of CIA mice were significantly decreased, and the number of osteoclasts in bone cavity reduced by treatment of triptohde. X-ray showed that the surface of bone of each articular was eroded, the joint space was narrow, BMD values of lumbar and knee were decreased compared with normal mice. Triptohde could significantly inhibit the damage of bones and joints of CIA mice, and increase the BMD values of lumbar and knee. Conclusion Triptolide can inhibit the damage of bones and joints of CIA mice.
