摘要
采用免疫组化法和DNA缺口末端标记法观察Rb抑癌蛋白、增殖细胞核抗原(PCNA)和凋亡细胞在宫颈癌变过程中的表达。结果发现Rb抑癌蛋白在正常宫颈上皮和不典型增生上皮均全部阳性表达,在宫颈癌组织中阳性率为57.7%,Rb基因突变随临床分期升高而递增,Rb阳性组的凋亡指数(AI)和凋亡指数/增殖指数(AI/PI)与阴性组无显著差异。凋亡指数与病理分级呈负相关(P<0.05)。证实Rb基因突变发生在宫颈癌的晚期,推测该基因不参与宫颈癌组织中细胞凋亡的调节。AI将成为新的预后指标。
Using immunohistochemical staining and DNA nick end labelling method, the expressions of Rb antioncogene protein , proliferating cell nuclear antigen (PCNA) and apoptotic cells were observed for their role in carcinogenesis of cervical cancer. It was shown that Rb protein was expressed in all normal cervix and cervical dysplasia. Rb was expressed in 57.7% of cervical carcinoma and mutation of Rb increased with increasing clinical stages. There was no difference of AI (apoptosis index) and AI/PI ratio between Rb positive tumors and negative ones ( P >0.05).Apoptotic index was negatively associated with the pathological types. It was demonstrated that Rb mutation seems to happen in the late stage of cervical cancer and that Rb was seemingly not to be involved in regulation of apoptosis in cervical carcinoma. AI may be a new prognostic factor.
