大鼠骨髓间充质干细胞的培养及鉴定

CULTURE AND IDENTIFICATION OF BONE MARROW MESENCHYMAL STEM CELLS IN RAT

摘要 目的:探讨大鼠骨髓间充质干细胞(MSC)体外分离培养及鉴定。方法:应用贴壁筛选法分离培养W istar大鼠髓间充质干细胞,用直接荧光抗体染色法对培养的细胞进行鉴定。结果:体外培养的原代MSC 9d融合,荧光抗体染色CD44、CD105表达阳性,CD14、CD34表达阴性。结论:贴壁筛选法在体外很容易分离培养和扩增MSC,可作为获得MSC的一种有效手段。 Objective:To investigate the methods of isolation,culture and identification of bone marrow mesenchymal stem cell(MSC)in rat.Methods:Rat bone marrow was obtained.The MSC were isolated and purified by using direct anchoring and identified by using direct fluorescence staining.Results:Primary cultured rat MSC reached fusion within 9 days.These cells were positive for CD44,CD105,negative for CD14,CD34 by fluorescence staining.Conclusion:Rat MSC can be isolated,cultured and amplified easily by using direct anchoring,which can be used as an effective method for culturing rat MSC.
