ACEI和ARB治疗改善糖尿病大鼠胰岛素分泌功能的研究 被引量:6

摘要 目的:研究血管紧张素转换酶抑制剂ACEI和血管紧张素II受体拮抗剂ARB对高脂喂养链脲佐菌素(STZ)诱导的糖尿病大鼠胰岛素和胰高糖素分泌及氧化应激功能的影响。方法:应用Enalaprial和Losartan对高脂喂养STZ诱导的糖尿病大鼠进行干预,放免法测定胰岛素和胰高糖素,ELISA检查氧化应激指标8-isoPGF-2α水平。并通过静脉葡萄糖耐量实验了解ACEI和ARB治疗对一相胰岛素分泌的影响。结果:ACEI或ARB治疗可以减少氧化应激,保护一相胰岛素分泌。结论:ACEI或ARB治疗对维持正常的胰岛素分泌可能起到了重要保护作用,从而降低血糖和减少糖尿病的发生。这可能是大规模临床研究中ACEI和ARB使血糖回归至正常水平或减少新发的糖尿病的发生率的深层机制之一。 Objective To study the effect of ACEI/ARB therapy on the secretion of insulin and glucagon as well as serum lipid peroxidation marker 8-iso PGF-2α levels in streptozoticin(STZ) induced diabetic rat models.Methods Twenty-four rat models of STZ induced diabetes were prepared(random blood sugar〉16.7mmol/L).Of which,8 models were fed enalaprial 5mg/kg/d,8 models were fed losartan 10μg/kg/d and 8 models left unterated.Fasting serum insulin,glucagon(with RIA) and 8-iso PGF-2α(with ELISA) levels were measured in these models and 8 control rats three weeks later.Intravenous glucose tolerance test(IVGTT) were performed in 12 rats(3 animals in each group) six weeks later.Results Serum levels of insulin in the treated models were higher than those in the non-treated models but without significance(P〉0.05).Serum levels of glucagon and 8-iso PGF-2α levels in the treated models were significantly lower than those in the non-treated models(P〈0.05).With IVGTT,the deficient insulin peak after glucose load was corrected to some degree(34×vs control〉6×) in the treated models.Conclusion ACEI/ARB treatment could improve the secretion of insulin in rat models of diabetes,which might be beneficial for controlling the progression of the disease.This phenomenon is consistent with the result of clinical study.
