摘要
为观察氯前列醇钠(PG-Cl)对卵巢癌细胞(SKOV3)增殖和凋亡的影响,应用细胞计数法测量癌细胞生长曲线,四甲基偶氮唑蓝(MTT)比色法测定细胞体外增殖情况,吉姆萨染色法细胞观察形态学,Ho-echest-33342凋亡染色法检测加药后细胞凋亡情况。结果显示,PG-Cl对SKOV3细胞抑制率的大小与药物浓度和时间成正比,在高浓度药物作用下效果显著。而在150μg/mL PG-Cl作用12 h后细胞发生凋亡,荧光显示核质固缩呈圆形或椭圆形团块。结果表明,PG-Cl对SKOV3细胞体外生长有显著抑制作用,并诱导细胞发生凋亡。
The purpose of this study was to investigate the cellular effects of cloprostenol sodium(PG-Cl) on the growth of SKOV3 cell lines of human ovarian cancer in vitro. The antitumor effect was studied using MTT assay, Giemsa staining assay, boold-cell counter system and hoechest-33324 cell staining assay. The results showed that the proliferation of SKOV3 cells was inhibited by PG-Cl in a dose and time dependent fashion, and was significant under high concentration. While cell apoptosis happened after 12 h in 150μg/mL concentration. The fluorescent assay showed round and oval nuclei. It is speculated that PG-Cl
