摘要
目的研究羟基喜树碱对膀胱癌细胞的作用,探讨其抑制膀胱癌的机制。方法将不同浓度的羟基喜树碱作用于人T24膀胱癌细胞,利用流式细胞仪、透射电子显微镜、荧光染色技术及DNA分析,对所作用的T24细胞进行动态分析以及细胞形态变化的观察。结果0.005mg/L的羟基喜树碱,主要是通过阻止T24细胞由S期进入G2/M期,并不诱导人T24膀胱癌细胞的凋亡;0.01~0.10mg/L时,凋亡的细胞数随着其浓度增加及作用时间的延长而增加。羟基喜树碱能够诱导人T24膀胱癌细胞的凋亡。结论羟基喜树碱作用机制之一是诱导人T24膀胱癌细胞的凋亡。
Objective To study the emechanisms of 10 Hydroxycamptothecine (HPT) inhibiting human urinary bladder cancer.Methods Using flow cytometry, electron microscopy and fluorescence microscopy, we studied the effects of HPT on human T 24 urinary bladder cancer cells. Results HPT at 0.005 mg/L inhibited T 24 bladder cancer cells by preventing the cells from S phase to G2/M phase and did not induce apoptosis. The apoptotic cells were induced by HPT at the concentration of 0.01~0.10 mg/L.Conclusion HPT can inhibit human T 24 urinary bladder cancer cells and induced apoptosis at some concentrations. The induction of apoptosis is a very important mechanism of 10 Hydroxycamptothecine to treat urinary bladder cancer cells.
