摘要
目的研究人参二醇组皂甙(PDS)对大鼠大脑皮层神经元膜L-型钙通道单通道电活动的影响。方法急性分离大鼠皮层神经元和细胞贴附膜片钳技术。结果PDS(1.5gL-1)可明显缩短大鼠皮层神经元L-型钙通道的平均开放时间,延长其平均关闭时间,降低其开放概率,而对该通道的电流幅值无明显影响,其作用与经典钙通道阻断剂vera-pamil相似但较弱。结论PDS对大鼠皮层神经元L-型通道有明显阻滞作用。
AIM To study the effects of panaxadiol saponins (PDS) on the single channel activity of Ltype calcium channel in cortical neurons of rats. METHODS Using patch clamp technique of cellattached configuration and acutely isolated cortical neurons of wistar rats. RESULTS PDS (15 g·L-1) shortened the open time of Ltype calcium channel, prolonged the close time, decreased the openstate probability, but exerted no effects on the amplitude of ionic current flow through the Ca2+ channel. The actions of PDS were similar to those of verapamil, a potent calcium channel blocker, but the effects of PDS were less than those of verapamil. CONCLUSION PDS has the blockade effects on the Ltype calcium channel in cortical neurons of rats.
