摘要 目的研究人参二醇组皂甙(PDS)对大鼠大脑皮层神经元膜L-型钙通道单通道电活动的影响。方法急性分离大鼠皮层神经元和细胞贴附膜片钳技术。结果PDS(1.5gL-1)可明显缩短大鼠皮层神经元L-型钙通道的平均开放时间,延长其平均关闭时间,降低其开放概率,而对该通道的电流幅值无明显影响,其作用与经典钙通道阻断剂vera-pamil相似但较弱。结论PDS对大鼠皮层神经元L-型通道有明显阻滞作用。 AIM To study the effects of panaxadiol saponins (PDS) on the single channel activity of Ltype calcium channel in cortical neurons of rats. METHODS Using patch clamp technique of cellattached configuration and acutely isolated cortical neurons of wistar rats. RESULTS PDS (15 g·L-1) shortened the open time of Ltype calcium channel, prolonged the close time, decreased the openstate probability, but exerted no effects on the amplitude of ionic current flow through the Ca2+ channel. The actions of PDS were similar to those of verapamil, a potent calcium channel blocker, but the effects of PDS were less than those of verapamil. CONCLUSION PDS has the blockade effects on the Ltype calcium channel in cortical neurons of rats.
