摘要
将编码禽流感病毒H5N1亚型HA基因(H5HA)的cDNA片段与CaMV35S启动子融合,通过农杆菌介导法转化马铃薯.分别用PCR,RT-PCR和Western斑点杂交方法对重组H5HA基因在马铃薯植株中的表达情况进行分析.实验结果表明,获得的12株转基因植株中,11株为阳性.Western斑点杂交检测表明H5HA重组蛋白已在马铃薯体内表达.这一结果将为利用马铃薯作为生物反应器生产禽流感口服疫苗提供理论依据.
In this study,we explore the possibility of H5HA gene expression in potato plant. The H5HA-encoding cDNA fragment is fused to the sequence of CaMV35S promoter. One potato cultivar(cv. Desiree) is used for Agrobacterium-mediadate transformation with pBIN19/35S-H5HA. Twelve transgenic,rooting shoots are obtained and multiplied. The expression of H5HA gene in transgenic plants is analyzed by PCR,RT-PCR and Western dot-blotting. The results show that H5HA gene is successfully expressed in the eleven transfomed potato plants,suggesting the possibility of using potato as a bioreactor for production of oral avian flu vaccine.
