摘要
[目的]枯草芽孢杆菌纤溶酶是一种碱性丝氨酸蛋白水解酶,具有强烈的纤溶活性,临床上可用于预防和治疗血栓栓塞性疾病,可用于开发新型溶栓药物。[方法]该研究采用PCR法克隆枯草芽孢杆菌纤溶酶基因及其前导肽序列,通过农杆菌介导转化野生型拟南芥,并利用纤维蛋白平板法检测BSFE纤溶活性。[结果]通过农杆菌介导转化获得转BSFE基因拟南芥,PCR检测证实BSFE基因已在转基因拟南芥基因组内整合,纤溶活性检测进一步证实BSFE可在转基因拟南芥体内表达,并可通过组织及根系分泌型表达,建立植物组织及根系分泌表达外源蛋白的系统模型。[结论]该研究为进一步阐明外源蛋白分泌表达机理及建立植物根系分泌生物反应器提供依据。
[ Objective] Bacillus subtilis fibrinolytie enzyme(BSFE) is one kind of alkaline serine proteinase, which can be applied clinically as a new type thrombolysis drug to prevent or cure the diseases related to thrombus, so it will be a promising market prospect. [Method] BSFE gene and its signal peptide were cloned by PCR and introduced into wild-type Arabidopsis by agro-bacterium mediated transfonrkation in this paper. And BSFE fibrinolysis activity was assayed on the plates with swine fthrin. [ Result] Transgenie Arabidopsis was obtained by Agrobacterium mediated transformation and BSFE gene integrated with Arabidopsis genome. The transgenie Arabidopsis could secret target protein from leaf, stem or root. And a model of expression and secretion foreign protein by plant tissue and root system were established. [ Conclusion ] The study laid the foundation for illuminating the mechanism of expression and secretion foreign protein by root system and establishing the bioreactor of plant's root system.
