摘要
目的研究人工合成杀菌肽(shiva-l)对直肠癌细胞HR8348的选择性杀伤作用机制。方法与结果用3H-标记胸腺脱氧核苷掺入法,证明shiva-1能抑制HR8348细胞的生长及DNA的合成,用激光共焦显微镜观察到HR8348细胞内的钙离子浓度较正常猴肾细胞(CV1)有明显降低。shiva-1选择性杀伤作用,首先攻击癌细胞的细胞膜,继而破坏细胞器,最后使细胞内容物逸出而死亡。结论人工合成杀菌肽shiva-1可能作为人的有效抗癌药物。
Objective Study on the selective cytocidal effect on adenocarioma cell of rectum HR8348 by synthetic cecropin shiva-l. Methods and Results The synthetic cecropin shiva-l inhibited the growth and DNA synthiesis of HR8348 cell by 3H-labelled thymidine deoxyribonucleoside corporation. Obervation under confucal scanning microscopy showed the intracellular caclicum ion of HR8348 Cell was significently decreased compared with the normal renal cell of monkey (cvi).The selective cytocidal effect of shiva-l attecked first the cell membrane, then destroyed the organelles, the intracellular contents poured out of the cell and died at last. Conclusion These results suggest that the synthetic cecropin shiva-l may be effective therapeutic at for human cancinoma.
