氯胺酮的抗惊厥作用及对兔定量脑电图β、θ频段的影响 被引量:6

Anticonvulsant effect of ketamine on β,θ-band of quantitative electroencephalogram in rabbits

摘要 目的用定量脑电图学方法探讨氯胺酮对士的宁惊厥的治疗作用。方法清醒家兔40只随机分成4组,氯胺酮1.25、2.5和5.0mg·kg-1组和对照组,每组10只。耳缘静脉注射士的宁0.25mg·kg-1制作惊厥模型,分别静脉注射氯胺酮或相应剂量的生理盐水,观察氯胺酮对家兔的死亡率和强直持续期的影响。连续监测脑电图,分析定量脑电图β、θ频段功率谱的变化。结果与对照组相比,氯胺酮1.25、2.5和5.0mg·kg-1组家兔惊厥强直持续期明显缩短(r=-0.8696,P<0.05),死亡率降低。在给药后1~5min,对照组的脑电图β频段明显增多(占80%以上),θ频段波形减少。与对照组相比,氯胺酮各剂量组在给药后1~5min所有观察脑区的β频段功率百分比均降低到50%以下,而大部分脑区θ频段功率百分比在给药后2~5min均提高1倍以上。结论氯胺酮能增加慢波频率,抑制痫性波发放,从而控制士的宁诱发的惊厥发作。 AIM To study the therapeutic effects of ketamine on strychnine-induced convulsion by quantitative electroencephalogram (QEEG) in rabbits. METHODS Forty awake rabbits were randomly divided into ketamine 1.25, 2.5, 5.0 mg·kg^-1 group and control group (n =10). Convulsion was induced by intravenous strychnine (0.25 mg·kg^-1). Normal saline and the different dose of ketamine were intravenously administered, respectively. Mortality and duration of tonic state were observed. The EEG activity was monitored continuatively. The percentage power of β, θ-band of QEEG sample was processed with the power spectral analysis in different time. RESULTS Compared with the control group, the duration of tonic state in ketamine 1.25, 2.5 and 5.0 mg· kg^-1 groups was shortened significantly in a dose-dependent manner (r = -0.869 6, P 〈 0.05) , and the mortality decreased. Within 1 - 5 min after administration, β-band power increased obviously and θ-band power decreased in EEG of the control group. Compared with the control group, every percentage of β-band power in all brain areas decreased to below 50% (P 〈 0.01 ) within 1 - 5 min after administration and the percentage of θ-band power in most areas increased above one time within 2 - 5 min after administration in ketamine groups. CONCLUSION Ketamine could increase the frequency of slow wave and inhibit firing in β-band to control convulsive paroxysm induced by strychnine.
