摘要
目的:建立适合混合蛋白质分离分析的高效毛细管电泳方法。方法:采用高效毛细管电泳中自由区带电泳分离模式,以标准蛋白质细胞色素C与溶菌酶为分析对象,对影响电泳分离的因素进行了考察。结果与结论:通过减少蛋白质管壁吸附、优选缓冲液体系及定性定量信息校正,使细胞色素C与溶菌酶获得了理想的分离,同时成功地应用于磷脂酶A2的优化分离与纯度分析,为蛋白质类生化药物的质量控制提供了简便、快速。
Objective: To establish a high performance capillary electrophoresis (HPCE) method for optimal separation of proteinaceous products. Methods: Cytochrome C and lysozyme were selected as model samples ,and the some factors affecting electrophoretic separation were investigated, based on capillary zone electrophoresis(CZE) mode. Results and Conclusion:Three combined strategies for the optimal separation of cytochrome C and lysozyme were described: the reduction of protein wall adsorption, the optimization for electrophoretic buffer system, and the normalization of HPCE qualitative and quantitative information.Successful application to phospholipase A2 purity analysis was demonstrated.The conclusion was obvious that HPCE should provide a simple, quick and effective analytical tool for proteinaceous drugs quality control.
