竹黄多糖液态发酵培养基优化研究 被引量:1

摘要 [目的]为竹黄多糖的工业化生产提供理论依据。[方法]通过组织分离,从野生竹黄子实体中获得能产生竹黄多糖的无性株,然后用其进行液态发酵培养,通过摇瓶正交试验确定其最佳培养基配方。[结果]碳源、氮源、生长因子和初始pH值的影响均达显著水平(α=0.05),其显著性依次为:碳源浓度>生长因子浓度>酸碱度>氮源浓度,在交互作用中只有CGF×CN的影响达显著水平。方差分析结果表明,最佳培养基配方为:蔗糖20 g/L+酵母膏8 g/L+玉米浆2 g/L,最佳初始pH值为6.0。当摇瓶装量为50/250 ml(V/V),接种量为10%(V/V),培养温度为28℃,摇床转速为120 r/min时,120 h后发酵液的竹黄多糖含量为8 g/L。[结论]该试验初步研究了利用竹黄无性菌株通过液态发酵生产竹黄多糖的培养基组成及培养条件。 [ Objective ] The purpose was to supply theoretical foundation for the industrialized production of Shiraia bambusicola polysaccharide. [ Method ] The asexual produeing strains of S. bambusicola polysaceharide were obtained from the fruiting bodies of wild S. bambusicola through tissue isolation. Then they were eultured in liquid medium by fermentation and their optimum medium formula was ascertained through orthogonal shaking flask experiment. [ Result] The influences of carbon source, nitrogen source, growth factors and initial pH value reached significant level and their siguifieances were as follows: eoncn, of carbon source 〉eonen. of growth factors 〉 pH 〉eonen. of nitrogen source. Among interactions, only the influence of CGF × CN reached significant level. The results of variance analysis showed that the optimum medium formula was sucrose 20 g/L + yeast paste 8 g/L + corn syrup 2 g/L and the optimum initial pH value was 6.0. When the eultare broth quantity was 50/250 ml (V/V), the inoculation amount was 10% (V/V), the culture temperature was 28 ℃ and the rotation speed was 120 r/min, the content of S. bambusicola polysaeeharide in the fermentation broth after 120 h was 8 g/L. [ Conclusion ] The medium composition and culture condition of producing S. bambusicola pelysaeeharide through liquid fermentation with asexual bamboo parasitic fungus were studied preliminarily in this experiment.
