竹红菌甲素(Hypocrellin A,HA)合并照光,对HeLa细胞有明显的杀伤作用。可使红细胞产生溶血和K^+释放。使红细胞膜ATP酶和乙酰胆碱脂酶失活。使肝癌细胞线粒体和微粒体膜脂质过氧化作用增加。体内应用HA合并照光,使S-180小鼠肿瘤产生光敏反应,但持续时间不长,一定时间后肿瘤复发。S-180小鼠静注[^(99m)TC】—HA,t1/2α,5.5分钟,t1/2β,231分钟。以肝脏内HA浓度最高,依次为脾、肺、肾、瘤、心、皮肤、肌肉、胃和脑。静注HA后第6小时,光敏反应最显著,24小时后光敏反应基本消失。HA合并照光,使肝癌细胞DNA断链,DNA修复缓慢,50分钟内未完全修复。
Hypocrellin A (HA) exerts a strong photodynamic effect on HeLa cells in the light, also causes rad cell hemolysis and K+ release. The red cell membrance ATPase and Acetylcholinesterse were inactivated. The lipid peroxidation of mitochondrial and micrcsomal membrane of hepatoma cell was enhanced. Moderate photodynamic action on S-180 tumors was observed, but it could not cure them. Intravenous injection of (99Tc)-HA into S-180 bearing mouse,t-α 5.5 min, t-β 231 min, the highest concentration was found liver and the lowest concentration was found in brain. The most obvious photodynamic action was observed at 6 hrs after HA administration, and disappeared at24hrs. HA in light caused marked DNA breakdown in hepatoma cells, and DNA repair was slow, the damaged DNA was not completely re-dpaired within 50 min.
