摘要
目的研究大鼠大脑中动脉(MCA)短暂闭塞后血脑屏障(BBB)通透性与脑毛细血管内皮细胞(BCECs)凋亡。方法大鼠MCA经腔内导入单线闭塞不同时间后再灌流24h,血管源性脑水肿(VBE)的程度用分光光度法测定Evans蓝(EB)渗出量评价,凋亡细胞用原位末端标记法观察。结果EB外渗量、凋亡的BCECs数显著高于对侧,随缺血时间的延长而增加,EB外渗量与凋亡的BCECs数显著相关。
Objective To examine the bloodbrain barrier (BBB) permeability and the number of apoptotic brain capillary endothelial cells (BCECs) after temporary middle cerebral artery (MCA) occlusion (MCAO) in rats.Methods MCAO was induced by cannulation with a monofilament nylon suture.The BBB permeability was assessed by quantitation of the extravasation of Evans blue dye (EB) by means of optical densitometry following formamide extraction.Results Both the extravasation of EB and the number of apoptotic BCECs increased with time of focal cerebral ischemia.The extravasation of EB in the ischemic hemisphere correlated well with the number of apoptotic BCECs in a coronal.Conclusion The apoptosis of BCECs may be involved in the mechanism of the BBB disruption.
