孕期锰染毒对子鼠神经行为的影响 被引量:11

摘要 目的:研究妊娠期锰染毒对子代神经行为的影响。方法:选用转棒试验、被动回避性跳台实验和Moris水迷宫试验,观察妊娠期大鼠锰染毒,其子鼠1月龄和2月龄时神经行为的改变,同时检测了子鼠脑内多巴胺含量和中脑内锰的含量。结果:①7.5mg和15mg染锰组的子鼠,其2月龄时的神经行为与正常组之间有着显著性的差异,其脑内多巴胺含量也明显低于正常对照组。②出生后2d、1月龄和2月龄染锰组子鼠中脑锰含量显著高于正常组。结论:母体内过多的锰可通过胎盘进入胎儿体内并引起子代神经行为的改变,其机理与多巴胺神经递质的减少,脑锰含量的增高有关。 Objective:Effects of prenatal exposure to manganese(Mn)on the behavior in offspring of rats were studied.Methods:Wistar pregnant rats were exposed to manganese chloride during gestation(0,3 75,7 5,and 15 mg Mn/kg i.p.).The behavioral alternations of offspring were observed at age of one and two months.Rota Rod Test,Step Down Test and Morris water Maze test were Carried as neurobehavioral test of offspring.The content of manganese in brain was examined by using Inductively Coupled Plasma Emission Spectrometry(ICP AES).Brain dopamine concentrations were measured by fluorescent analysis.All data were analyzed by Statistical Package for the Social Science(SPSS/PC +).Results:①neurobehavior of offspring rats which thein mother were exposed to 7 5 and 15 mg/kg were dsignificantly different from control group at age of two months,and the brain dopamine concentration decreased at age of two months.②The content of manganese in midbrain increased at 2 day,one month and two months of age in the offspring rats.Conclusion:It suggested that maternal exposed to excessive Mn can transfer through the placenta to fetal and resulted in neuro behavoaral alternation,decreasing of brain dopamin in offsping at age of two months. Prenatal animals are suscepted to Mn neurotoxicity in this expervinent.
