环氧化酶-2及细胞色素芳香化酶基因mRNA在育龄妇女子宫内膜息肉中的表达 被引量:11

Expressions of cyclooxygenase-2 mRNA and aromatase cytochrome P450 mRNA in endometrial polyps among women of reproductive age

摘要 目的研究环氧化酶-2(COX-2)基因和细胞色素芳香化酶P450(P450 arom)基因在育龄妇女子宫内膜息肉(EP)中的表达及相关性。方法应用逆转录聚合酶链反应(RT-PCR)技术,检测15例正常子宫内膜、30例育龄妇女EP及息肉周围内膜中COX-2 mRNA和P450arom mRNA的表达,并进行相关性分析。结果COX-2mRNA和P450arom mRNA在EP及息肉周围内膜中均有较强表达。EP中COX-2和P450 arom的表达与息肉周围内膜相比均无显著性差异(P均>0.05)。在EP中,P450 arom和COX-2 mRNA表达与月经周期无关(P均>0.05)。在息肉周围内膜中,P450 arom mRNA和COX-2 mR-NA在分泌期的表达均明显高于增生期(P均<0.01)。P450 arom在EP及息肉周围内膜中的表达与COX-2呈正相关。结论COX-2和P450 arom与育龄妇女EP的发病相关,二者协同作用促进EP的发生与发展。 Objective It is to study the mRNA expressions of cytochrome aromatase P450 (P450 arom) and cyclcoxygenase-2 (COX- 2) in endometrial polyps (EP) among women of reproductive age and their correlation with the diseaseand their correlation. Methods The expressions of COX - 2 mRNA and P450 arom mRNA were assessed by RT - PCR in 15 cases of normal endometrium, 30 cases of EP and the endometriurn around EP. The correlated analysis was done. Results The COX - 2 and P450 arom mRNA expressed in EP and the endometrium around EP were strong. No significant changes of COX - 2 mRNA and P450 arom mRNA expressions were found in EP and the endometrium around EP (P 〉0.05). In EP, no significant changes of COX - 2 and P450 arom mRNA expressions were found throughout the menstrual cycle. The expressions of COX - 2 and P450 arom mRNA of secretory phase were significantly higher than that of proliferative phase in the endometrium around EP (P 〈 0.01 ). There was a positive correlation between P450 arom mRNA expression and COX - 2 mRNA expression in EP. Conclusion COX - 2 and P450 arom mRNA expressions are closely correlated with the pathogenesis of EP among women of childbearing age. The synergistic action of the two enhances the occurence and development of EP.
