摘要
GP2是从人胃癌组织中提取的硫粘蛋白相关的糖蛋白抗原,本研究以抗GP2单克隆抗体AH5通过免疫组化技术检测GP2在胃癌、结肠癌、直肠癌、肝癌、肺癌及其它肿瘤组织中的表达。结果显示,GP2在胃癌、结肠癌和直肠癌中有高水平的表达,其阳性表达率分别为84%、72.7%和77.8%,而在其它肿瘤组织中表达率较低(P<0.01),另外GP2在胃肠肿瘤中不同病理类型,其阳性表达率和表达强度不同,其中腺癌的阳性表达率最高,而高分化者的阳性表达强度高于低分化者。研究结果提示,硫粘蛋白相关抗原GP2在胃肠道肿瘤中有特异性高表达,可以作为一种独立的胃肠道肿瘤及其癌前病变的肿瘤标志物。
GP2 is a sulfomucin associated glycoprotein antigen extracted from human gastric cancer specimen. The present work was aimed on the immunohistochemical determination of the expression of GP2 in the cancer tissues of stomach, colorectum, liver, lung etc, using anti GP2 monoclonal antibody AH5.The result shows that the GP2 showed a high level expression in gastric, colonic and rectal cancers with the positive rates of 84%, 72.7% and 77.8% respectively, whereas the expression of GP2 is of low level in other kinds of tumor ( P <0.01).Besides, the positive rate and intensity of the expression of GP2 varied in different gastrointestinal tumors of variable pathological types. Being highest in malignant adenoma and in undifferentiated ones. The result of our study led to the conclusion that the sulfomucin associated GP2 has a distinctive high expression in gastrointestinal tumors, and thus can be taken as a special tumor marker of the gastrointestinal tumor and might herald early carcinogenesis.Therefore, it is very helpful in the diagnosis of gastrointestinal tumors,and identification of gastrointestinal cancer metastasis as well as the carcinogenetic process in gastrointestinal tissues.
