摘要
报告1株鼠抗人胃癌单克隆抗体(单抗)MGd1相应抗原的初步研究。单抗MGd1经饱和硫酸铵(50％浓度)盐折沉淀及DEAE-52纤维素柱层析纯化后,与溴化氢话化的Sepharose4B偶联制备免疫吸附剂。用单抗免疫亲和层析的方法从胃癌组织可溶性抗原提取液中纯化出MGd1相应抗原。免疫酶斑点试验、TLC放射免疫自显影检测提示MGd1相应抗原既溶于PBS缓冲液又易溶于甲醇氯仿液中,高效薄层色谱、SDS-PAGE及免疫印迹等实验分析证实,MGd1相应抗原含有中性糖脂糖蛋白两种成分,抗原决定簇存在于糖链上。Mgd1-Ag是一种新的具有深入研究价值的胃癌相关抗原。
The MGd was purified by passing through a DEAE-52 column,and covalently coupled with CNBr-activated sepharose 4B.By means of immunoaffinity chromatography,the MGdl corresponding antigen was purified from gastric cancer tissue.The results of high performance thin layer chromatography,TLC immunoautoradiography,SDS-PAGE and immunoblotting studies confirmed that the MGdl corresponding antigen was a neutral glycolipid and glycoprotein.It is a new gastric cancer associated antigen.
