摘要
用人工合成心肌线粒体ADP/ATP载体(ANT)多肽免疫大鼠,动态观察末次免疫后第14、28、42天时大鼠血清抗体滴度及心功能变化,免疫电镜胶体金技术观察免疫复合物在心、肝、肾的沉积.结果:ANT多肽免疫组大鼠中有63%(17/27)产生了特异性抗体,心功能发生了抗体滴度相关性的降低,心肌纤维及线粒体上可见特异性免疫复合物沉积.对照组大鼠未产生特异性抗体,心、肝、肾中均无免疫复合物沉积.
Twenty-seven rats were immuned with the artificially synthesized antigenic determinants of cardiac ADP/ATP carrier (ANT). Antibody titers in sera were dynamically determined ,and cardiac functions and immunocomplex depositions on heat,liver and kidney were observed at day,14,28 and 42 after the latest immunization. The results showed that the titers of specific antibody in sera were obviously increased in 63% of the 27 rats immuned with ANT peptide. And simultaneously cardiac function impairment was occurred and closely correlated with the levels of antibody titers. Immunocomplexes were observed to be deposited specifi-cally on cardiac myocytes. It was concluded that specific cardiac impairment can be induced by the immunization with the antigenic determinant of ANT selected .which is similar to the whole ANT protein. Synthetic peptide of ANT can partly replace the whole ANT protein in the further study of autoantibodies against ANT.
