摘要
采用不同大豆品种的下胚轴、上胚轴、小真叶和幼胚作外植体进行植株再生的研究。结果表明汾豆33号诱导植株再生率最高;诱导频率依次为下胚轴>上胚轴>小真叶>幼胚;下胚轴在B5+NAA0.3mg/L+KT1mg/L培养基上可诱导出愈伤组织并直接分化成苗,植株再生频率为34%,最高达50%。获得大豆高频率再生植株,收到较饱满的种子。
Various explants, hypocotyl, epicotyl, euphyllum, and germinating embryo of four soybean cultivars were used for regeneration study. It was revealed that cultivar Fendou 33 produced the highest regeneration rate. Regeneration rates of various explants were, in the order from high to low, hypocothl, epicothl, euphyllum and germinating embryo. The plantlet regeneration rate for Fenodou 33′hypocothle on the B5 medium with 0.3 mg/L+1mg KT/l was 34% and the highest plate up to 50%. Mature seeds were obtained from the regenerated plants. The study lay certain foundation for further inducing somatic variation and making genetic transformation.
