摘要
目的建立人脑间变性星形细胞瘤的裸小鼠原位移植模型,并探讨移植瘤细胞的生长状况。方法将表达表皮生长因子受体(EGFR)的新鲜人脑间变性星形细胞瘤的手术标本移植于裸小鼠右尾状核并传代,观察各代移植瘤的生长特性及组织学特点。结果移植瘤在裸小鼠右尾状核已连续传至7代,向周围鼠脑侵袭性生长;瘤细胞排列紊乱,核大深染且异型明显;移植瘤中EGFR的表达率为70%。结论本法建立的人脑间变性星形细胞瘤的原位模型能够真实地模拟其原发脑肿瘤的生物学特性,为进一步对间变性星形细胞瘤的放、化疗等实验研究提供可靠的动物模型。
Objective To establish an orthotopic transplantation model of malignant glioma of human brain in nude mice,and analysize its biological features. Methods Fresh anaplastic astrocytoma masses with expression of EGFR from hmnan brain were collected and orthotopicly transplanted into the fight caudate nucleus of nude mice. The newly formed xenografts were sharply minced into pieces and re - transplanted. HE staining and MRI scanning were performed to investigate the histopathological and imaging features of the xenografts. The expression of EGFR in xenografts was also investigated by immunohistochemical assays of EnVision. Results Histopathologically, the cerebral xenografts were poorly differentiated anaplastic astrocytoma with invasiveness to normal brain tissue. MRI revealed a poor - delimitated mass with surrounding tissues, which perfectly recapitulated imaging features of the original tumor. Expression of EGFR was positive in the cerebral xenografts with the ratio of 70%. Conclusions The orthotopic xenografts of brain anaplastic astrocytoma could faithfully recapitulate the biological features of the original tumor clinically and pathologically, and provides a reliable animal model not only for studying the biological features but also for experimental treatment of brain malignant glioma
