摘要
目的构建针对胶质瘤发生、发展分子机制研究的组织芯片。方法根据研究目的设计118例样本(包括各级别人胶质瘤组织、恶性胶质瘤细胞系多细胞球体、胶质瘤干细胞球体以及用于对照的正常人脑组织、人胚胎脑组织、神经干细胞球体)的阵列,应用组织芯片仪制作组织芯片蜡块,切片后进行HE染色和光镜观察。结果构建的组织芯片蜡块包含118个点阵;熔蜡后有个别点阵发生移位,切片后无点阵缺失;HE染色观察符合原始病理诊断。结论针对胶质瘤发生、发展分子机制研究而制作的组织芯片是胶质瘤分子病因研究的有用工具。
Objective To construct a tissue microarray for the study of gliomas aiming at the molecular mechanisms of occurrence and development. Methods A microarray containing 118 specimens ( including all grades of human glioma tissues, malignant glioma cell line multicellular spheroids, glioma stem cellular spheroids, and containing normal human brain tissues, human fetal brain tissues and neural stem cellular spheroids for the use of control ) was designed according to the purpose of study, and a tissue arrayer was used for the construction of the tissue microarray paraffin block, then performing HE staining and observed by microscope after section. Results The tissue microarray paraffin block constructed contained 118 arrays, few of which were observed shifting after melting, and no array lost after section; the view of the HE staining slide were coincide with the original pathologic diagnoses. Conclusion The tissue microarray, which was constructed for the study of gliomas aiming at the molecular mechanisms of occurrence and development, is a useful tool in the research of the molecular etiology of glioma.
