摘要
采用高效液相色谱(HPLC)法测定了凹叶景天中槲皮素和异鼠李素含量。采用Kromasil C18(250mm×4.6mm,5μm)色谱柱,以乙腈-水-磷酸(43:57:1)为流动相,流速为1.0mL·min^-1,检测波长为360nm。得到槲皮素回归方程为y=74197x+40608,R^2=0.9998,槲皮素在3.6~18.0μg·mL^-1范围内线性关系良好,平均加样回收率为100.65%,RSD=2.02%;异鼠李素回归方程为y=285344x+5535.1,R^2=0.9998,异鼠李素在1.06~5.30μg·mL^-1范围内线性关系良好,平均加样回收率为101.99%,RSD=4.95%。该方法简便、高效、准确度较高,结果令人满意,为制定凹叶景天的质量标准及指纹图谱的研究提供了依据。
The contents of quercetin and isorhamnetin in Sedum emarginatum Migo were determined by high performance liquid chromatography (HPLC) using Kromasil C18 (250 min × 4.6 mm, 5 μm) column,with acetonitrile : water : phosphoric acid (43 : 57 : 1) as mobile phase at a flow rate of 1.0 mL · min^-1 and detection wavelength of 360 nm. The results showed that the regression equation for quercetin was y = 74197x+ 40608, R^2 =0. 9998, quercetin of 3.6-18.0 μg · mL^-1 had a good linear relationship, the average recovery was 100.65%, RSD=2.02% the regression equation for isorhamnetin was y = 285344x 4-5535.1, R^2= 0. 9998, isorhamnetin of 1. 06- 5. 30μg · mL^-1 had a good linear relationship, the average recovery was 101.99 %, RSD =4.95 %. This method is simple, efficient with high accuracy and satisfactory results, so it can provide reference for the establishment of quality standard and fingerprint spectrum study of Sedum emarginatum Migo.
