摘要
目的:研究不同粒径磁性纳米FeOx在外加磁场作用下对肝癌细胞增殖、凋亡的影响。方法:采用共沉淀法制备得到不同粒径FeOx,将不同粒径FeOx吸附于细胞表面,并加载静磁场(SMF)及100 Hz交变磁场(EMF)照射。运用CCK-8检测纳米粒子吸附后在外加磁场作用下Bel-7402细胞增殖情况,运用流式细胞仪检测细胞凋亡、死亡率及细胞周期变化。结果:不同粒径磁性纳米FeOx在SMF照射时间低于72 h时能使Bel-7402细胞增殖速度增快,当照射时间大于72 h时,细胞增殖速度未发生明显变化但部分细胞发生凋亡,凋亡率为(3.1±0.78)%。而经EMF照射后37 nm FeOx吸附细胞增殖被抑制,细胞周期实验结果表明大部分细胞被阻滞在G0/G1期,细胞出现大量死亡与凋亡,凋亡率达到(23.34±3.6)%、死亡率达到(57.24±2.7)%。结论:纳米粒子未有外加磁场照射时不能对细胞产生明显的影响,外加SMF照射时,细胞的增殖及DNA代谢未发生明显的变化,但细胞发生凋亡;外加EMF照射时,细胞增殖被抑制并发生明显的凋亡及死亡,细胞DNA代谢明显紊乱,且具有较小粒径的磁性纳米FeOx在EMF作用下对肝癌细胞影响最为显著。
Objective: To study the effects of different diameters of nano FeOx powders on proliferation and DNA metabolism of heptoma-cell lines Bel-7402. Methods: Different diameters of nano FeOx powders were utilized to dope the heptoma-cell lines Bel-7402 through the co-precipitation meanings, then cells were exposed by static magnetic field(SMF) and extremely low frequency altering magnetic field(EMF) with 100 Hz. After exposed by external fields, CCK-8 kit was used to detect the effects on proliferation of cells caused by nano powders. Apoptosis kit and DNA cycle kit was used to detect the effects caused by nano FeOx of cell apoptosis, death and cell DNA cycle metabolism. Results: Different diameters nano FeOx could not influence the cell at all under SMF exposure but could influence the cells under EMF, when the diameter was 37 nm the highest apoptosis and death ration were (23.34 ± 3.6) % and ( 57.24 ± 2.7 ) %, respectively, and the most of cells were prohibited in G0/G1 period of DNA cycle. Conclusion: nano powders could not influence the cells at all without external magnetic field exposure. Under SMF exposure the proliferation and DNA metabolism of cells absorbing by nano powders were not influenced, but the apoptosis ratio of cells was increased. After exposure by EMF cells absorbing by nano 'powders could be affected in proliferation, apoptosis and DNA metabolism, furthermore, all the effects were more obvious in the cells absorbing by nano powders with little diameter under EMF exposure.
