沉默Bcl-2基因对A549细胞的影响

摘要 目的探讨Bcl-2基因在抑制细胞凋亡的作用。方法利用RNA干扰技术,通过向人非小细胞肺癌A549细胞中导入siRNA表达载体,抑制Bcl-2基因表达,诱导细胞凋亡。结果Bcl-2基因沉默导致A549细胞发生凋亡。结论Bcl-2家族成员之间相互拮抗的关系及由此产生的A549细胞凋亡,为治疗肿瘤提供了一条新的途径。 Objective To explore the suppressive effect of Bcl-2 gene on tumor cell apoptosis. Methods RNA interference technology was used to suppress Bcl-2 gene expression and induce cell apoptosis by introducting siRNA expression vector into A549 cell of human non-small cell lung cancer. Results Silencing of Bcl-2 gene induced A549 cell apoptosis. Conclusions Members of Bcl-2 family are mutually antagonistic, and further induce A549 cell apoptosis, which provides a new strategy to treat tumor.
