摘要 采用C18反相柱层析法分离大豆异黄酮糖苷,再将其水解制备成异黄酮苷元,研究其对结肠癌HT-29和肺癌A549细胞增殖和凋亡的影响。用MTT比色法观察大豆异黄酮对肿瘤细胞增殖的影响,用流式细胞术检测大豆异黄酮对细胞周期分布的影响,用TUNEL染色法检测其对HT-29细胞凋亡的影响。结果表明,大豆异黄酮呈时间和浓度依赖性地抑制肿瘤细胞增殖,诱导肿瘤细胞凋亡。同时改变结肠癌HT-29细胞周期分布,使其阻滞于G2/M期。提示,大豆异黄酮可通过改变细胞周期分布和诱导细胞凋亡发挥抗肿瘤作用。 Total glycosides were extracted from soybean hypocotyl, then isoflavone glycosides were isolated with C18 reversed phase column chromatography. Finally isoflavone aglycones were prepared by hydrolysis of isoflavone glycosides and their effects on proliferation and apoptosis of and lung carcinoma cell A549 were investigated. MTT assay was used to examine the proliferation of tumor cell, flow cytometry was used for analysis of cell cycle distribution, TUNEL method was used for analysis of cell apoptosis. The results showed that soybean isoflavones inhibited the proliferation of carcinoma cells in a time- and concentration-dependent manner and induced apoptosis. In addition, soybean isoflavones lead to colon carcinoma cell HT-29 G2/M arrest. It is suggested that soybean isoflavones could exert an anti-cancer effect via cell cycle arrest and induction of apoptosis.
