全反式维甲酸联合三羟异黄酮对肺癌细胞增殖及凋亡的影响 被引量:3

摘要 目的探讨全反式维甲酸(all-trans retinoic ac id,ATRA)联合三羟异黄酮(Gen iste in)抑制肺癌细胞的增殖和促进其凋亡的作用。方法采用不同剂量ATRA、Gen iste in及两者联合处理人肺腺癌细胞株A549,观察A549细胞生长情况并运用MTT法检测其生长抑制情况,流式细胞仪进行细胞周期分析及凋亡率检测。结果A549肺癌细胞经ATRA、Gen iste in及两者联合处理后,细胞增殖能力明显下降,更多的细胞被阻滞于G2/M期,细胞凋亡明显增加,其中以两者联合用药最为显著。结论ATRA联合Gen iste in能协同抑制肺癌A549细胞的恶性增殖并促进其凋亡。 Objective To investigate the effects of all-trans retinoic acid (ATRA) and/or Genistein on the proliferation and apoptosis of human pulmonary carcinoma cell line A549 in vitro. Methods The A549 cells were treated with ATRA at the dose of (5, 10, 50) × 10^-6 mol/L, and/or Genistein at the dose of ( 10, 20, 40) × 10^-6 mol/L for 1 -7 d. The proliferation of A549 was assessed by MTT and cytometry, and the rate of apoptosis was analyzed by flow cytometry. Results ATRA and Genistein combination could obviously inhibit the growth of A549 cells, arrested the cells in G2/M phase and induced more cell apoptosis. Conclusion Genistein could enhance the effect of ATRA on proliferative suppression and apoptosis induction of A549 cells.
