毛细管气相色谱法测定辛夷挥发油软胶囊中的α-蒎烯、β-蒎烯、柠檬烯和1,8-桉叶素

Determination of α-pinene,β-pinene,limonen,and 1,8-cineole in Flos Magnoliaevolatile oil soft capsula by capillary gas chromatography

摘要 目的建立测定辛夷挥发油软胶囊中α-蒎烯、β-蒎烯、柠檬烯和1,8-桉叶素的测定方法。方法采用毛细管气相色谱法测定。BP-5毛细管柱(30m×0.25mm,0.22μm);进样器温度180℃;FID检测器温度200℃;柱温采用程序升温;分流比1:30;苯乙酮为内标。结果α-蒎烯在0.054~2.505μg线性关系良好,平均加样回收率为98.6%,RSD为0.72%(n=9);β-蒎烯在0.070~3.204μg线性关系良好,平均加样回收率为99.0%,RsD为0.65%(n=9);柠檬烯在0.068~3.190μg线性关系良好,平均加样回收率为101.1%,RSD为0.56%(n=9);1,8-桉叶素在0.301~10.205μg线性关系良好,平均加样回收率为99.6%,RSD为1.24%(n=9)。结论该方法简便、准确、重现性好,可作为辛夷挥发油软胶囊的质量控制方法。 Objective To develop a method for determination of α-pinene, β-pinene, limonen, and 1, 8- cineole in FlosMagnoliae volatile oil soft capsula. Methods The contents of α-pinene, β-pinene, limonen, and 1, 8-cineole were determined at the same time by capillary gas chromatography with sequential increase of temperature programming. Capillary column BP-5 (30 m × 0.25 mm, 0. 22 μm) was used, injector temperature 180 ℃, flame ionization detector (FID) temperature 200 ℃, split ratio 1 : 30, acetophenone as internal standard. Results A good linear relationship of a-pinene was at a range of 0. 054--2. 505 μg, the average recovery was 98.6%, and RSD was 0.72 % (n=9); A good linear relationship of β-pinene was at a range of 0. 070--3. 204 μg, the average recovery was 99.0 %, and RSD was 0.65% (n=9) ; A good linear relationship of limonen was at a range of 0. 068--3. 190 μg, the average recovery was 101.1%, and RSD was 0.56% (n=9) ; A good linear relationship of 1, 8-cineole was at a range of 0. 301--10. 205 μg, the average recovery was 99. 6%, and RSD was 1.24% (n=9). Conclusion The method is simple and accurate one with good reproducibility and could be used for the quality control of Flos Magnoliae volatile oil soft capsula.
