摘要
目的监测小鼠心肌缺血/再灌注损伤(MIRI)过程中,血清炎性细胞因子TNF—α、IL-6水平,中性粒细胞活性及氧自由基含量的变化,以及瘦素(1eptin)预处理对其影响,探讨瘦素预处理在小鼠MIRI过程中的作用及其机制。方法24只雄性C57BL/6小鼠随机分为四组:①假手术组;②心肌缺血/再灌注模型组(MIRI组);③MIRI瘦素预处理组(leptin+MIRI组);④MIRI术后瘦素处理组(MIRI+leptin组)。在MIRI手术后6h,四组小鼠分别取标本,测定小鼠血清炎性细胞因子水平(TNF-α、IL-6)、髓过氧化物酶活性(MPO)水平、血清丙二醛(MDA)活性以及超氧化物歧化酶(SOD)含量。结果瘦素预处理可降低细胞因子TNF—α、IL-6的水平。降低小鼠MPO活性,减少MDA含量并升高SOD活性。结论瘦素预处理可减少中性粒细胞的聚集与浸润,减轻脂质过氧化和自由基损伤,对MIRI有保护作用。
Objective To investigate the mechanism of the effect of leptin pretreatment on myocardial isehemia reperfusion injury (MIRI)in mouse. Methods Twenty four male C_.57BL/6 mice were randomized into 4 group: ①the shamoperation group; ②the myocardial ischemia reperfusion group ( MIRI group ), established by blocking left coronary artery anterior descending branch for 30 rain followed by reperfusion;③the leptin pretreatment group (Leptin + MIRI group), established by injecting intraperitoneally leptin at 30 min before ischemia; ④the leptin post - treatment group ( MIRI + Leptin group ) ; established by injecting intraperitoneally leptin at 30 min after reperfusion. Blood and heart samples were harvested from each mouse at 6 hours after reperfusion. We then analyzed the samples for tumor necrosis factor - alpha ( TNF - α) and interleukin - 6 ( IL - 6 ) levels associated with MIRI, myeloperoxidase ( MPO ) level, malondialdehyde (MDA) contents and superoxide dismutase ( SOD ) activity. Results Administration of leptin before MIRI significantly reduced serum TNF - α and IL - 6 levels, decreased myeloperoxidase (MPO) level, improved malondialdehyde (MDA) content and restored superoxide dismutase ( SOD) activity. Conclusion Pretreatment of leptin showed protective effects on mouse MIRI by Ameliorating the up - regulation of leptin level in mouse MIRI model. Up - regulation of leptin level induced by MIRI deteriorates myocardial injury.
