摘要
目的建立黄芩药材高效液相色谱-质谱(HPLC-MS)总离子流指纹图谱的评价方法。方法SHIMADZU VP-ODS柱(250mm×4.6mm,5μm),乙腈与0.1%甲酸进行梯度洗脱;流速为0.8ml/min,检测波长276nm。结果得到分离度、重现性均较好的黄芩药材HPLC-ESI-MS总离子流色谱指纹图谱,标示了14个共有峰,以各峰的保留时间和相对峰面积作为数量化基础并借助SPSS13.0数据处理软件进行模糊聚类分析,结果表明山东产黄芩相似性较好,与外省样品有较明显区别。结论黄芩药材指纹图谱的构建,为控制其质量提供了依据。
Objective To establish an evaluation method of Scutellaria baicalensis by HPLC-MS total ionic chromatographic fingerprints. Methods The HPLC separation was performed on SHIMADZU VP-ODS column（250 mm ×4.6mm, 5 μm）, gradient eluted using acetonitrile and 0.1% formic acid-wate solvent system with the flow rate of 0.8 ml/min and detected at 276 nm. Results Perfect fingerprints were obtained which can be used for the evaluation of Scutellaria baicalensis ; 14 common peaks were confirmed in fingerprints. Clustering analysis showed an excellent similarity of Scutellaria baicalensis. Conclusion The method can be applied to establish an evaluation method and quality control of Scutellaria baicalensis.
