大鼠脑出血后低氧诱导因子表达及中药干预的作用 被引量:2

The expression of hypoxia inducible factor-1α and its regulation with traditional Chinese medicine in rats models of experimental intracerebral hemorrhage

摘要 目的:利用大鼠脑出血模型探讨脑出血期间脑组织中低氧诱导因子1α(HIF-1α)表达的时程和分布特征及中药干预对脑出血后HIF-1α表达的影响。方法:39只体重250—300g的健康成年雄性SD大鼠,采用Nath改良法建立脑出血模型,随机分为脑出血组、中药(脑血通)干预组,另设生理盐水和正常组作为对照。分别于造模后第6h、24h、72h、第7天,利用Bederson等的3级标准评定动物的神经系统功能,然后分别处死取脑用免疫组织化学法检测HIF-1α的表达。结果:正常脑组织和生理盐水组无HIF-1α的表达。脑出血后第6小时同侧基底核血肿区周围出现HIF-1α表达增加,至第72小时达高峰,第7天明显下降。与单纯脑出血组比较,中药干预后可以使上述各时间点HIF-1α表达较脑出血组减少,在第24h、72h、7d有显著性意义(P<0.01),神经功能障碍体征改善在第72h、第5天时间点有显著性意义(P<0.05)。结论:脑出血后血肿周围HIF-1α的表达明显上调,并可能参与了脑出血后血肿周围神经细胞的继发性损伤过程;中药具有神经保护作用,能有效地抑制脑出血后HIF-1α表达。 Objective: To investigate the changes of expressions of hypoxia-inducible factor-1α (HIF-1α) and its distribution in brain tissue in rats models of experimental intracerebral hemorrhage(ICH),and the possible effects of traditional Chinese medicine on the expressions of HIF-1α. Method: The rats'model was established by Nath modified method. Thirty-nine male Sprague-Dawley rats (weight 250--300g) were randomly divided into intracerebral hemorrhage group and intracerebral hemorrhage treated with traditional Chinese medicine(naoxuetong) group; saline group and normal group were negative controls. At the 6^th, 24^th,72^nd h and the 7^th d after operation, the rat's neurological functions were assessed with Bederson's grade. Then rats were sacrificed respestively, and the brain tissues were taken out. Immunohistochemistry analyses of expressions of HIF-1α were observed. Result: HIF-1α was undetectable in brain tissues at normal group and saline group. However, the positive HIF-1α expressions in neurons were detected in ipsiiateral basal ganglia around the hematoma region in intracerebral hemorrhage group, with up-regulation of HIF-1α expressions occurred as early as at the 6^th h, peaked at the 72^nd h, and then declined at the 7^th d. Compared with intracerebral hemorrhage group HIF-1α expressions down-regulated significantly in intracerebral hemorrhage treated with traditional Chinese medicine group at the 24^th h,72^nd h,7th d (P〈0.01),and neurological functions improved significantly at the 72^nd h,5^th d (P〈0.05). Conclusion: The expressions of HIF-1α up-regulated after experimental intracerebral hemorrhage around the hematowa in rats, which may contribute to the secondary cerebral injury process surrounding hematoma;the protective effects of traditional Chinese medicine may be related with inhibiting the expressions of HIF-1α.
