摘要
植物血凝素(PHA)可诱导再生障碍性贫血(AA)病人外周血及骨髓单个核细胞产生 CFU-GM 抑制活性,此活性与上清液中γ-干扰素(IFN-γ)的含量高度相关,并可被γ-干扰素单克隆抗体所中和。加入环孢菌素 A(CsA)(1μg/ml)能有效地抑制 IFN-γ的产生,从而消除其对 CFU-GM 的抑制,提示上述作用可能为 CsA 治疗 AA 的机理之一。
A CFU-GM-inhibiting activity was found in supernatants of aplastic anemia patients' PHA-induced peripheral blood mononuclear cells and bone marrow mononuclear cells.It was strongly correlated with the γ-IFN concentration and could be eliminated by anti-γ- IFN McAb.With presence of cyclosporin A,the γ-IFN produced by peripheral blood mo- nonuclear cells of the patients studied reduced significantly,followed by a marked decrease in CFU-GM-inhibiting activity in the supernatants just as caused by anti-γ-IFN McAb. All above indicate that cyclosporin A can induce the release of hematopoietic progenitor from γ-IFN-mediated suppression,which may be one of the mechanisms of cyclosporin A in tre- atment of aplastic anemia.
