原花青素对大鼠脑缺血再灌注损伤Caspase-3表达和神经元凋亡的影响

Effect of procyanidin on neural cell apoptosis and expression of Caspase-3 following cerebral ischemic reperfusion injury in rats

摘要 目的探讨原花青素(PC)对脑缺血再灌注损伤大鼠Caspase-3表达和神经元凋亡的影响。方法40只大鼠随机分为假手术组、缺血再灌注组、PC低剂量治疗组和PC高剂量治疗组,线栓法建立局灶性脑缺血再灌注模型。各组于缺血90min再灌注24h用免疫组化法观察大鼠额顶部皮质Caspase-3蛋白表达,采用TUNEL法检测神经细胞凋亡,并进行TTC染色和HE染色观察脑梗死体积及病理形态学变化。结果缺血再灌注组与假手术组比较,Caspase-3表达增加,凋亡细胞明显增加;PC治疗组与缺血再灌注组比较均显著减少Caspase-3表达,减少凋亡细胞,高、低剂量组之间差异有统计学意义(P〈0.05)。PC治疗组脑组织缺血损伤病理学改变均明显轻于缺血再灌注组,PC高剂量治疗组缺血改变亦轻于低剂量治疗组。PC治疗组脑梗死体积均较缺血再灌注组减小,且高、低剂量组之间差异具有统计学意义(P〈0.05)。结论PC对缺血再灌注脑损伤具有保护作用,其机制可能与通过减少Caspase-3表达,抗凋亡有关。 Objective To study the effect of procyanidin on neural cell apoptosis and the expression of Caspase-3 of cerebral ischemia reperfusion(I/R) injury in rats. Methods 40 rats were randomly divided into 4 groups, which were sham operated group, I/R group, low dose procyanidin treated group and high dose procyanidin treated group. The focal middle cerebral artery occlusion (MCAO) model was made by suture-occluded method. After MCAO for 90min following 24h of reperfusion, neural cell apoptosis and the expression of caspase-3 was investigated with TUNEL and immunohistochemistry. HE staining and TTC staining was also used. Result Compared with sham operated group, neural cell apoptosis rate and the expression of caspase-3 were increased at the 24th hour of reperfusion in the ischemic territory( P 〈 0. 05 ) . Compared with I/R group, low and high dose procyanidin treated group reduced expression of caspase-3 and neural cell apopto- sis rate in a dose-dependent manor ( P 〈 0. 05 ). The change of ischemie impairment in proeyanidin treated group was Jess than that of I/R group, and the change of high dose procyanidin treated group was less than that of low dose procyanidin treated group. Compared with that of I/R group, cerebra/ infarction volume of procyanidin treated group was decreased in a dose-dependent manor ( P 〈 0. 05 ). Conclusion Procyanidin may reduce cerebral ischemia-reperfusion injure by reducing expression of caspase-3 and neural cell apoptosis.
