摘要
目的①探讨红松松塔水溶性物质提取的最佳工艺;②探讨红松松塔水溶性物质对小鼠自然杀伤细胞(natural killing cells)活性的影响。方法①采用L9(34)正交实验设计法,以质量提取率为指标,优选红松松塔水溶性成分提取的最佳工艺;②用体外和体内给药的方式,采用乳酸脱氢酶(LDH)释放法考察红松松塔水溶性成分对小鼠自然杀伤细胞活性的影响。结果①提取的最佳工艺为:红松松塔的粉碎粒度为20目,提取时间为1h,提取次数为1次,料液比为1∶8;②红松松塔提取物经大孔吸附树脂分离后,经体内外实验研究发现:在体外实验中红松松塔水提物(包括总提物、水洗脱物和乙醇洗脱物)能提高小鼠自然杀伤细胞的活性;在体内实验中,红松松塔总提物和乙醇洗脱物均能明显提高小鼠自然杀伤细胞的活性,而水洗脱物没有此效果。结论红松松塔水溶性物质能够增强机体的自然杀伤细胞活性,即是能够增强机体的固有免疫力。
OBJECTIVE To search the optimal extraction technology of pinecone of Pinus koraiensis Sied.et Zucc (here simply termed PC). To study the effect of the extracts on mice natural killing cells. METHODS L9(3^4) orthogonal test was used to optimize the technology for the water-solubility extracts of PC, with the extraction ratio as the criteria. The activity of mice natural killing cells was measured by lactic dehydrogenase (LDH) release test after the cells were treated with extracts of PC in vitro and in vivo. RESULTS The optimum conditions for extracting the water-solubility substances of PC were that PC's particle size was of 20 screen mesh, 8 times of water, extracted once and each time lasted 1 h. After being separated by macroporous adsorptive resins, the extracts of PC increased the activity of mice natural killing cells in vitro. The total extracts and extracts eluted by 40% ethanol raised the activity of mice natural killing cells in vivo and substances eluted by water didn't show the activity. Meanwhile the activity was concentration-dependent. CONCLUSION The extraction technology was stable and accurate, and it was suitable for extracting water-solubility substances of PC. The water-solubility extracts of PC extracted by this technology could enhance the activity of mice natural killing cells.
