摘要 目的优化血凝和血凝抑制(HA/HI)试验条件,提高HA/HI试验的稳定性和准确性。方法在不同的缓冲液、pH值、猪红细胞浓度、BSA浓度下进行HA/HI试验,选择合适的HA/HI条件。结果pH7.0、0.1%BSA、0.2 mol/L PBS、1%猪红细胞可作为HA/HI试验合适的反应条件,猪血球可保存12 d不影响试验结果。结论方法优化后稳定性增强、检测时间缩短、准确性提高。 Objective To optimize the conditions of hemagglufination and hemagglutination inhibition assay and improve it's stability and accuracy. Methods HA and HI titers were detected under different solutions, pH values, BSA concentrations and porcine erythrocytes concentrations. The condition making it more sensitive or stabilize was selected as the optimum. Results 0.2 mol/L PBS containing 0.1% BSA at pH7.0 was confermed the best buffer for dilutting virus and preparing 1% porcine erythrocytes. Porcine erythrocytes conserved in Alsever' s solution at 4℃ within 12 days could keep HA/HI assays as sensitive as the fresh porcine erythroeytes. Conclusions The improved assay was more accurate and stabilize. It's detection time was shortened after optimization.
