甘蓝型油菜种子黑色素提取条件的优化 被引量:5

摘要 [目的]探讨甘蓝型油菜种子黑色素提取的最佳条件。[方法]以中双5号甘蓝型油菜籽为材料,用碱溶解-酸沉淀法提取甘蓝型油菜种子黑色素,通过单因素试验探讨提取剂NaOH浓度(1%、2%、3%、4%)、提取温度(40、60、80、100℃)、提取时间(1、2、3、4 h)、提取次数(1、2、3、4次)、沉淀pH值(1、2、3、4)对甘蓝型油菜种子黑色素提取率的影响,并采用正交试验优化提取条件。[结果]5种因素对甘蓝型油菜种子黑色素提取率均有不同程度的影响。[结论]甘蓝型油菜种子黑色素的最佳提取条件是:NaOH浓度为3%,提取温度为80℃,提取时间为1 h,提取次数为2次,沉淀pH值为4,其提取率为13.07%。 [ Objective ] The study aimed to discuss the optimum condition for extracting the melanin from the rapeseeds of Brassica napus L. [ Method ] With the rapeseeds of B. napus Zhangshuang 5 as the tested material, its melanin was extracted by alkaline dissolution-acid deposition method. The single factor experiment was designed to explore the effect of NaOH concn.( 1%, 2%, 3%, 4% ), extraction temperature (40, 60, 80, 100 ℃ ), extraction time ( 1, 2, 3, 4 h), extraction times ( 1,2, 3,4 times) and deposition pH value ( 1, 2, 3, 4) on the extraction of melanin from rapeseeds of B. napus. The extraction conditions were optimized through the orthogonal test. [ Result] Five single factor had different effects on on the extraction of melanin from rapeseeds of B. napus. [ Conclusion ] The optimum condition for extracting the melanin from the rapeseeds ofB. napus was follows as 3% NaOH, extraction temperature of 80 ℃, extraction time of 1 h, extraction times of 2 times, pH value of 4. Under this condition, the yield of melanin reached to 13.07%.
