摘要
[目的]为灵芝多糖的工业化生产提供理论指导。[方法]以赤灵芝子实体为材料,采用超声波法提取其中的灵芝多糖,并通过单因素和正交试验研究超声功率、超声时间、提取温度、料液比对灵芝多糖得率的影响,确定灵芝多糖的最佳提取工艺。[结果]单因素试验结果表明,提取温度为50℃,超声时间为40 min时灵芝多糖得率最高,当超声功率小于500 W时,灵芝多糖得率随超声功率的增大快速增加。各因素对灵芝多糖得率的影响由大到小依次为:超声功率>料液比>提取温度>超声时间;灵芝多糖的最佳提取工艺为:超声功率500 W、提取温度45℃、超声时间35 min、料液比1∶25。[结论]在最佳工艺条件下,灵芝多糖的得率为2.75%。
[ Objective] The study aims to provide the theoretical guidance for industrial production of polysaccharide from Ganoderma lucidum. [ Method ] With fruiting bodies of G. lucidum as the materials, the polysaccharide from G. lucidum was extracted by ultrasonic assistant method, and the effects of ultrasonic power, ultrasonic time, extraction temperature and solid-liquid ratio on yield rate of polysaccharide from G. lucidum were studied with single factor and orthogonal tests And then the optimum extraction technology for polysaccharide from G. lucidum was determined. [ Result] The results of single factor test showed that the yield rate of polysaccharide from G. lucidum was the highest when extraction temperature was 50 ℃ and ultrasonic time was 40 min. When ultrasonic power was lower than 500 W, the yield rate of polysaccharide from G. lucidum was rapidly increased with the increase of ultrasonic power. The effects of factors on yield rate of polysaccharide from G. lucidum from big to small in order was ultrasonic power 〉 solid-liquid ratio 〉 extraction temperature 〉 ultrasonic time, and the optimum extraction technology for polysaccharide from G. lucidum was ultrasonic power of 500 W, extraction temperature of 45 ℃, ultrasonic time of 35 min, solid-liquid ratio of 1:25. [ Conclusion ] Under the optimum extraction technology condition, the yield rate of polysaccharide from G. lucidum was 2.75%.
