摘要
采用试管法、纸层析法(PPC)和薄层层析法(TLC),初步鉴定了山核桃蒲壳中的氨基酸、多糖、黄酮、皂苷、挥发油、强心苷、生物碱组分.采用正交实验法,考察提取温度、乙醇浓度、液料比、提取时间对总生物碱提取率的影响.研究结果表明,提取温度与乙醇浓度对总生物碱提取率的影响显著(P<0.05),较佳提取条件为:50%乙醇,60℃水浴提取2次,每次1.5h,液料比5∶1.
An experiment was conducted to study on the chemical constituents in peels of Carya cathayensis Sarg. by three methods: test tube method, PPC method and TLC method. And the chemical constituents such as amino acid, polysaccharide, flavonoid, saponin, volatile oil, cardiac glycoside, and alkaloid were identifided. Effects of extraction temperature, ethanol concentration, ratio of solvent to sample, and extraction hours on the extraction yield of total alkaloids were analyzed by the orthogonal test. The results indicated that extraction temperature and ethanol concentration had significant effects on the extraction yield of total alkaloids (P〈0.05). And the optimum extraction conditions were 50% ethanol, heating for 1.5 h at 60 ℃ water bath for two times, ratio of solvent to sample 5:1.
