原位PCR检测肝组织中HCV基因状况分析

摘要 为了探讨肝组织中HCVRNA的意义,使用原位PCR检测技术,对26例血清HCV标志物阳性肝病者进行检测。结果显示该方法特异性强,肝组织HCVRNA阳性率为692%,阳性信号主要定位在细胞浆,个别呈浆核型,但仍以浆型为主。肝功损害与肝细胞中HCVRNA有一定相关性。 HCV RNA in 26 hepatic biopsy' specimens with positive anti-HCV or/and HCV RNA in sera were detected by situ polymerase chain reaction to probe the significance of HCV RNA in the hepatic tissue.The result demonstrated the technique had an excellent specificity,and the positive rate of HCV RNA was 69 2%(18/26) in the specimens studied,83 3%(15/18) of the positive HCV RNA markers was seen in hepatic cytoplasm,16 7(3/18) HCV RNA markers located between cytoplasm and nuclei(cytoplasm-nuclei type).Hepatocytes with positive of HCV RNA signal was less than 5% in 11 cases, 5% to 25% in 6 cases,and 26% to 50% in only one case.No HCV RNA marker in the hepatic tissues was seen in cases with positive HBV but negative HCV,or both negative HBV and HCV in the sera.
