摘要
目的观察血管紧张素Ⅱ(AngⅡ)对体外培养的肾小管上皮细胞生长的影响。方法用不同浓度(10-6、10-7、10-8、10-9mol/L)AngⅡ刺激体外培养的肾小管上皮细胞,在不同的时间点(24、48、72h)用四甲基偶氮唑盐(MTT)法检测细胞增殖;用不同浓度(10-6、10-7、10-8mol/L)AngⅡ刺激体外培养的肾小管上皮细胞,作用48h后,流式细胞仪检测细胞周期。结果MTT法检测结果显示AngⅡ作用于肾小管上皮细胞后,吸光度(A)值明显增加(P<0.05),10-6mol/LAngⅡ作用后上升最明显(P<0.01),且在48h作用点达到峰值。流式细胞仪结果显示AngⅡ能使肾小管上皮细胞在S期比例增加,DNA合成增加。结论AngⅡ刺激肾小管上皮细胞增殖,48h增殖作用最显著,并在一定范围内呈浓度依赖性。
Objective To investigate the effects of angiotensin Ⅱ(Ang Ⅱ ) on proliferation of human renal tubular epithelial cell. Methods Cultured human renal tubular epithelial cells were treated with Ang Ⅱ at different concentrations (10^- 9, 10^- 8 , 10^- 7 and 10^- 6 mol/L respectively) for different time (24 h,48 h,72 h). The proliferation of renal tubular epithelial cells was determined by MTT colorimetry. Cell cycles of the cell were detected by flow cytometer. Results MTT increased after the human renal tubular cells were treated with AngⅡ , especially at the concentration of 10^-6 mot/L (P〈0.01) for 48 h. The cells in the S-stage increased, also at the concertration of 10^- 6 mol/L. Conclusion It is suggested that AngⅡ stimulates the proliferation of human renal tubular epithelial cells in a dose-dependent pattern, and at the time of 48 h, the effect was the most significant (P〈 0.01).
